Progesterone in Oil Injection Instructions

  1. Necessary materials:
    • One 10 cc (ml) vial of Progesterone in oil
    • One 3 cc syringe with attached needle (gray cap) (for medication draw)
    • One 25 gauge 11/2-inch needle (blue cap) (for injection)
    • Alcohol swabs

  2. Snap off the cap from the Progesterone bottle
  3. Make sure the needle attached to the 3 cc syringe is secured on tightly
  4. Turn Progesterone bottle upside down, and using the 3 cc syringe with attached needle draw up your daily dose of progesterone (0.5 cc, 1 cc, 1.5 cc, or 2 cc as prescribed by your physician)
  5. Remove the 22 gauge needle (gray cap) from the 3 cc syringe and replace with the 25 gauge 11/2-inch injection needle (blue cap)
  6. It is recommended that you numb the injection site with ice 5 minutes prior to injection
  7. Just prior to injection, rub 3 cc syringe (containing drawn up Progesterone) with attached injection needle (capped) rapidly between both hands in a rolling motion to warm medication (heat warms and thins oil, making injection easier)
  8. To remove any air bubbles, point the needle up and gently tap on the syringe until all the bubbles rise to the top, then push the plunger carefully until a small drop of liquid begins to appear from the tip of the needle
  9. Wipe skin with alcohol swab at intended injection site
  10. Remove cap off of needle and administer medication intramuscularly:
    • The injection should be given in the upper outer quadrant of the buttocks (see diagram)
    • Pinch skin at injection site into a mound between thumb and forefinger and hold the syringe like a pencil with the other hand
    • Push the needle through skin into muscle at a 90-degree angle until needle disappears
    • Gently pull back on plunger until a small air bubble appears in the syringe (if blood appears in the syringe, do not panic, pull the needle out a bit, change direction, push back all the way in until needle disappears under skin), then inject medication
    • Pull needle out of skin and wipe injection site with an alcohol swab, rubbing the site gently (this will help the absorption of the medication); occasionally a small amount of blood will be visible, which is not a reason for any concern
  11. Massage injection site for two to five minutes after injection, you may apply a heating pad to the injection site (will facilitate intramuscular absorption)
  12. Maintain unused progesterone at room temperature for future injections

Possible skin/subcutaneous reaction
: A red area may appear around the injection site and/or the injection site might itch and/or bruising may occur. These are all excepted reactions and will disappear over time. Due to the viscous nature of progesterone in oil, moderate induration (thickening) of the subcutaneous tissue may occur. This is normal and is of no cause for concern. If the indurated area becomes very tender and/or warm to touch or if redness/bruising around the injection site appear to be enlarging or spreading please notify us immediately.

Storage: Progesterone in oil bottle should be kept at room temperature.

Use the disposable syringe(s) only once. Dispose of syringe(s) properly in a biohazard container. When biohazard container is full return it to our office or your pharmacy. Never leave syringes, drugs, or needles where children can reach them. Keep careful track of your prescribed dose of medication as it may change throughout the course of your treatment cycle. Whenever uncertain about your injection, contact our office for clarification.
